The Advantages of Fluorescent Proteins over Luciferase for In Vivo Imaging

چکیده

What are Fluorescent Proteins and Luciferase? Fluorescent proteins (FP) and luciferase are two categories of proteins that glow. Fluorescent proteins include the green fluorescent protein (GFP) series which was originally extracted from jelly fish and the cyan fluorescent protein series (CFP) which was extracted from corals. Many different versions of fluorescent proteins had been engineered by mutating the original proteins. Their glow mechanism is fluorescence which means these proteins first absorb energy of one color light (excitation), digest the energy and then emits the partially consumed energy as a different color light.

A New Reporter Gene Technology: Opportunities and Perspectives

The paper summarizes the current status of the reporter gene technology and their basics. Reporter gene technology is widely used to monitor cellular events associated with gene expression and signal transduction. Based upon the splicing of transcriptional control elements to a variety of reporter genes, it “reports” the effects of a cascade of signaling events on gene expression inside cells. ...

A novel luciferase fusion protein for highly sensitive optical imaging: from single-cell analysis to in vivo whole-body bioluminescence imaging

Fluorescence and bioluminescence imaging have different advantages and disadvantages depending on the application. Bioluminescence imaging is now the most sensitive optical technique for tracking cells, promoter activity studies, or for longitudinal in vivo preclinical studies. Far-red and near-infrared fluorescence imaging have the advantage of being suitable for both ex vivo and in vivo analy...

Use of Gfp for in Vivo Imaging: Concepts and Misconceptions

Although GFP and fluorescent proteins are used extensively for in vivo imaging, there are many misconceptions about GFP imaging especially compared to luciferase. GFP is not toxic, indeed, transgenic animals with GFP expressed in every cell (1) live as long as non-transgenic animals. Cancer cells with GFP are as aggressive and malignant as the cells without GFP (2-4). Cell lines can be made ver...

Comparison of human optimized bacterial luciferase, firefly luciferase, and green fluorescent protein for continuous imaging of cell culture and animal models.

Bioluminescent and fluorescent reporter systems have enabled the rapid and continued growth of the optical imaging field over the last two decades. Of particular interest has been noninvasive signal detection from mammalian tissues under both cell culture and whole animal settings. Here we report on the advantages and limitations of imaging using a recently introduced bacterial luciferase (lux)...

Investigation of an Optimized Context for the Expression of GFP as a Reporter Gene in Chlamydomonas Reinhardtii

Background: Chlamydomonas reinhardtii is a novel recombinant eukaryotic expression system with many advantages including fast growth rate, rapid scalability, absence of human pathogens and the ability to fold and assemble complex proteins accurately, however, obstacle relatively low expression level necessitates optimizing foreign gene expression in this system. The Green Fluorescent Protein (G...
